چکیده

Purpose: To investigate the action of miR-483-3p in pediatric pneumonia and identify potential biomarkers.
Methods: Bronchoalveolar lavage was collected from 38 patients 25 healthy children. The expression determined using qRT-PCR. Alveolar macrophages children were cultured used for functional assays. Transwell assay conducted to evaluate macrophage cell migration. Cell viability apoptosis evaluated lipopolysaccharide (LPS)-induced human pulmonary alveolar epithelial cells (HPAEpiCs) by CCK8 (cell counting kit - 8) or flow cytometry, respectively.
Results: MiR-483-3p significantly elevated bronchoalveolar patients, when compared (p < 0.001). MiR-483-3p, which targets insulin-like growth factor 1 (IGF1), decreased mRNA protein IGF1 reduced motility macrophages. counteracted LPS- induced decrease increase HPAEpiCs. Conditioned medium transfected with inhibitor increased cellular LPS-induced However, conditioned mimics apoptosis.
Conclusion: negatively regulates promote progression pneumonia, providing a therapeutic target pneumonia.

Oncogenic role of miR-483-3p at the IGF2/483 locus.

hsa-mir-483 is located within intron 2 of the IGF2 locus. We found that the mature microRNA (miRNA) miR-483-3p is overexpressed in 100% of Wilms' tumors. In addition, colon, breast, and liver cancers exhibit high or even extremely high levels of miR-483-3p in approximately 30% of the cases. A coregulation with IGF2 mRNA was detected, although some tumors exhibited high expression of miR-483-3p ...

Circulating miR-483-3p and miR-21 is highly expressed in plasma of pancreatic cancer

Several recent studies have revealed that microRNAs (miRNAs) have a role in carcinogenesis and cancer development, and that it is stably detectable in plasma/serum. The aim of this study was to test whether miR‑483-3p as well as miR‑21 could be plasma biomarkers for PDAC. The plasma samples were obtained from three groups including 32 pancreatic ductal adenocarcinoma (PDAC) patients, 12 patient...

Over-expression of the miR-483-3p overcomes the miR-145/TP53 pro-apoptotic loop in hepatocellular carcinoma

The miR-145-5p, which induces TP53-dependent apoptosis, is down-regulated in several tumors, including hepatocellular carcinomas (HCCs), but some HCCs show physiological expression of this miR. Here we demonstrate that in HCC cells carrying wild-type TP53 the steady activation of the miR-145 signaling selects clones resistant to apoptosis via up-regulation of the oncogenic miR-483-3p. Expressio...

miR-125a-3p and miR-483-5p promote adipogenesis via suppressing the RhoA/ROCK1/ERK1/2 pathway in multiple symmetric lipomatosis

Multiple symmetric lipomatosis (MSL) is a rare disease characterized by symmetric and abnormal distribution of subcutaneous adipose tissue (SAT); however, the etiology is largely unknown. We report here that miR-125a-3p and miR-483-5p are upregulated in the SAT of MSL patients, promoting adipogenesis through suppressing the RhoA/ROCK1/ERK1/2 pathway. TaqMan microRNA (miR) array analysis reveale...

IGF2-derived miR-483 mediated oncofunction by suppressing DLC-1 and associated with colorectal cancer

Emerging evidence indicates that IGF2 plays an important role in various human malignancies, including colorectal cancer (CRC). Hsa-miR-483 is located within intron 7 of the IGF2 locus. However, the mechanism by which increased IGF2 induces carcinogenesis remains largely elusive. DLC-1 has been identified as a candidate tumor suppressor. In this study, we aimed at investigating whether miR-483 ...
